肺癌组织中ToPoⅡ表达与癌细胞体外药敏反应关系的研究

摘　　要：目的:探讨肺癌组织中ToPoⅡ与癌细胞体外药敏反应的关系。方法:收集本院2005年至2006年手术切除的肺癌组织标本80例,免疫组织细胞化学方法测定其组织中ToPoⅡ表达状态,用MTT法检测ToPoⅡ不同功能状态下各种肿瘤细胞对化疗药物Vp-16(足叶乙甙,ToPoⅡ抑制剂)的敏感性及差异。结果:以Vp-16的IC50值计算,ToPoⅡ高表达组患者是低表达组患者的1/6,显示出较高的化疗敏感性。结论:肺癌组织中ToPoⅡ功能状态可能对其化疗的敏感性有影响。Objective： To explore the relationship between the expression of ToPo Ⅱ in lung cancer and their sensitivity to etoposide in vitro. Methods： The expression of ToPo Ⅱ in 80 patients with lung cancer was examined by immunohistochemistry SP method. Meanwhile, the sensitivity to etoposide in different ToPo Ⅱ expression level was tested by MTT. Results： The sensitivity to etoposide was significantly different in different expression levels of ToPo Ⅱ in lung cancer. The IC50 of the Vp - 16 in higher expression group was significantly lower than that in lower expression group. Conclusion： The ToPo Ⅱ function status in lung cancer directly influences their sensitivity to etoposide.
