摘 要:目的:观察川芎油口服给药对脑缺血再灌注大鼠脑组织ICAM-1、血浆TNF-α及ET的影响。方法:线栓法建立大鼠局灶性脑缺血再灌注损伤(MCAO)模型,脑缺血2小时、再灌注22小时后采用放射免疫方法测定血浆中内皮素(ET)和肿瘤坏死因子-α(TNF-α)的含量,采用免疫组化法测定大鼠脑组织细胞间黏附因子1(ICAM-1)的表达。结果:川芎油能降低血浆中ET、TNF-α含量以及脑组织中ICAM-1的表达。结论:川芎油通过降低脑缺血再灌注损伤大鼠血浆中ET、TNF-α含量以及脑组织中ICAM-1的表达,减轻脑缺血再灌注损伤时的炎症反应以发挥其脑组织保护作用。Objective: To observe the possible mechanism of volatile oil of Lingusticum chuanxiong Hort(LCH) to the rats during focal cerebral ischemia-reperfusion injury. Methods: Transient middle cerebral artery (MCA) occlusion was induced as described by Longa, After 2 hours of cerebral ischemia and 22 hours of reperfusion , tumor necrosis factor-α( TNF -α) , endothelin (ET) levels in serum were observed by radioimmunoassay methods and intercellular adhesion molecule-1 (ICAM-1) were observed by immunohistochemical methods. Results: Compared with MCAO model group, level of the tumor necrosis factor-α (TNF - α) , endothelin (ET) and intercellular adhesion molecule-1 ( ICAM-1 ) levels were lower significantly in the groups treated with volatile oil. Conclusion : The volatile oil of Ligusticum chuanxiong Hort (LCH) could protect brain damage after focal cerebral isehemia reperfusion in vivo, and the possible mechanism seemed to be that it could release the injury of the inflammatory reaction.
