壳聚糖膜对成骨细胞的毒性研究

摘　　要：目的:根据细胞毒性评价的试验方法,探索自制的一种壳聚糖膜对成骨细胞的毒性作用。方法:选取原代培养的胎鼠颅骨成骨细胞,用改良MTT法检测壳聚糖膜对成骨细胞的毒性作用,试验设计有壳聚糖膜的成骨细胞实验组、阴性对照组和阳性对照组,以第0.5、1、2、3、5天为检测时间点。结果:壳聚糖膜对成骨细胞各时间点的毒性分级为0级或1级。结论:该材料有良好的生物相容性。Objective ：To evaluate in vitro cytotoxicity of a chitosan film on osteoblasts. Methods： Osteoblasts derived from fetal rat calvaria were cultured on chitosan films. Cell proliferation ability was tested by MTT Assay. The relative growth rate was calculated and the cytotoxicity was graded. Results：The relative growth rates of the osteoblast exposed directly to chitosan films were above 100% at the 0.5, 2, 3 and 5 d; cytotoxicity grade was 0. For day 1, cell relative growth rate was over 90%, and the cytotoxicity grade was 1. There was no statistical difference between the absorptance of chitosan film group and the negative control group（ P 〉 0.05 ）. Conclusion：Cytotoxicity of this chitosan film is grade 0 or 1, which satisfies the safety standard. This chitosan film has good biocompatibility.
