摘 要:目的探讨缝隙连接蛋白/细胞间缝隙连接通讯(CX/GJIC)下调对急性肝功能衰竭(AHF)大鼠肝细胞增生修复的影响。方法大鼠190只(SPF级,江西实验动物质量检测中心提供),随机分成苯巴比妥组PB(n=60),对照组(n=30),模型组(n=100)。PB组动物诱导后与模型组采用四氯化碳腹腔注射法造模,造模后1、3、7、10、14d采取肝组织标本行免疫组化查Cx32、切开标记荧光染料传输(IL/DT)查GJIC功能。结果(1)造模后动物立即出现精神萎靡,活动及摄食减少,尿黄,严重者多脏器出血,昏迷甚至死亡。PB与模型组的组间病死率差异显著(P<0.01)。(2)免疫组化显示,模型组的Cx32蛋白主要集中在胞膜上,细胞间,数量较PB组明显增多,且随着时间呈进行性增加的趋势;PB组的Cx32主要分布在胞浆内,数量极少。(3)荧光染料传输试验发现,模型组的染料传输能力在造模后与对照组比明显减低(P<0.01);PB组染料传输能力明显受抑制,除在第3天与模型组间的差别无统计学意义外,其余时间点与其它各组间差异均显著(P<0.05)。结论通过预先下调肝细胞间的CX/GJIC,可以减轻AHF急性期的肝脏损害,促进残存肝细胞增生和肝功能的好转,降低AHF大鼠的病死率。Objective To investigate whether down-regulate of CX/GJIC reduces liver damage in rats with acute hepatic failure (AHF). Methods One hundred and ninety male Wistar rats were random divided into three groups: PB Group (n=60), AHF Group (n=100) and control Group (n=30). Acute hepatic failure was induced by intraperitoneal injection of carbon tetrachloride (CCL4) in olive oil(V:V=1:1). Prior to injection rats in PB Group were fed with 800PPM phenol barbital (BP) in drinking water for a week. Liver specimens were taken at d1,3,7,10 and 14. Expression of Cx32 protein in liver tissue was examined by immunohistology and CX/GJIC was detected by incision Loading/Dye-Transfer Method(IL/DT). Results The mortality of PB Group was lower than that of AHF group (P〈0.01).The expression of Cx32 in GP group was mainly located in cytoplasm and lower than that in other groups (P〈0.01).The dyes transfer as demonstrated by IL/DT method was inhibited in all animals with induced AHF, especially in PB group (P〈0.01). Conclusion Down-regulation of CX/GJIC may reduce liver injury in AHF rats.
