摘　　要：目的构建Survivin靶向SiRNA重组表达载体。方法根据GeneBank数据库提供的Survivin基因所有变异体的共同核苷酸序列,选择设计3对带发夹状结构的核苷酸序列,克隆到空载体pGensil-1中,转化DH5α菌株,提取质粒进行酶切鉴定和测序分析。结果经酶切鉴定证实Survivin靶向SiRNA重组表达载体测序结果与设计完全一致。结论Survivin靶向SiRNA重组表达载体构建成功。Objective To construct SiRNA recombinant expression vector targeting survivin gene. Methods Genome sequences of survivin gene was retrieved from gene bank, three pairs of oligonucleotides were synthesized and inserted into plasmid pGensil-1, DH5α strains were transformed, plasma was extracted and identified by restriction endonuclease and sequence analysis. Results It was confirmed by restriction endonuclease that SiRNA recombinant expression vector targeting survivin gene was coincided with the designs completely. Conclusions The SiRNA recombinant expression vector targeting survivin gene has been constructed successfully.
