摘  要:采用PCR-DGGE技术,用两对引物对湖泊沉积物细菌群落结构进行了比较研究.结果表明:沉积物样品中细菌群落结构显示了明显的空间分布多样性差异,随着沉积深度的增加,细菌群落多样性指数降低.采用不同引物扩增的产物DGGE图谱聚类相近,均分成0~20cm和20~45cm两大类,但引物341f/534r得出的相似性Cs值在30.2%~75.9%之间,引物341f/926r得出的相似性Cs值在34.3%~84.1%之间,采用不同引物不同沉积深度样品群落结构之间的相似性和优势种动态变化存在差异.The bacterial community of lake sediment samples were comparatively analyzed with two sets of universal primers by PCR-DGGE, GE. The results indicated that the successional shifts in community structure were more evident at greater sediment. The Shannon-Weiner index (H) decreased along with the depth of sediment. Principle coordinate analysis suggested that the bacterial communities along the sediment core with different primers could be separated into two same groups which located 0-20 cm and 21-45 cm respectively. On the basis of Dice Coefficient (Cs), similarity analysis of the DGGE band patterns indicated that the composition of bacterial community was discrepancy at different layers of the sediment samples, Cs between the top and the bottom layers were 30.2%- 75.9% with the primers of 341f/534r and 34. 30%- 84.1% with the primers of 341f/926r respectively. The community diversity and dominant species dynamics were obviously different based on the different sets of universal primers by DGGE.
