摘 要:目的:观察苦碟子口服液(KDZOS)对实验性高脂蛋白血症大鼠血清总胆固醇、脂蛋白-胆固醇代谢的影响及其抗氧化作用。方法:将60只Wistar雄性大鼠随机分成6组,每组10只。空白对照组,给予普通饲料,其余为高脂饮食组,给予高脂饲料,建立实验性高脂蛋白血症模型,KDZOS按10、20和40g.kg-1.d-1给大鼠连续灌胃30d,测定血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白-胆固醇(HDL-C)及低密度脂蛋白-胆固醇(LDL-C)含量,计算动脉粥样硬化指数(AI)。同时测定血浆血栓素A2(TXA2)和前列环素(PGI2)水平,血清和肝脏丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性以及全血粘度,并观察肝脏脂肪沉积情况。结果:与高脂模型组比较,KDZOS20和40g.kg-1组大鼠的TC、TG、LDL-C、TC/HDL-C、LDL-C/HDL-C、AI、TXA2、MDA及全血低切、中切、高切黏度明显降低(P<0.05或P<0.01),而HDL-C、PGI2、PGI2/TXA2及SOD明显提高(P<0.05或P<0.01),病理检查可见肝脏的脂肪沉积明显减轻。结论:KDZOS能调节实验性高脂蛋白血症大鼠的血脂代谢,抑制肝脏脂肪沉积。Objective To observe the effect of Kudiezi Oral Solution (KDZOS) on total cholesterol (TC), lipoprotein cholesterol metabolism and antioxidative activity in experimental hyperlipoproteinemia rats. Methods 60 Wistar rats were divided into 6 groups and each group had 10 rats. Control group was given normal food. The models of experimental hyperlipoproteinemia were established by giving lipid-riched feeds to Wistar rats for 15 d. The contents of TC, triglyceride (TG), high density lipoprotein eholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) were measured and the atherogenic index (AI) was calculated. At the same time, the prostacycline (PGI2) and thromboxane (TXA2) levels in plasma, the malondialdehyde (MDA) contents and the superoxide dismutase (SOD) activities in serum and liver, and the viscosity of whole blood were also measured respectively in hyperlipidemia rats. In addition, the fat accumulation in liver was observed. Results In rats treated with KDZOS (with doses of 20 and 40g·kg^-1 i. g for 30 d), the levels of TC, TG, LDL-C, TC/HDL-C, LDL-C/ HDL-C, AI, TXA2, MDA and the viscosity of whole blood were decreased significantly (P〈0.05 or P〈0.01), HDL-C, PGI2, PGI2/TXA2 and SOD were increased significantly (P〈0.05 or P〈0.01), meanwhile the fatty degeneration of hepatic cells was relieved. Conclusion KDZOS can adjust the blood lipid metabolism in hyperlipoproteinemia rats and restrain fatty deposition in liver.
