山仙颗粒对Lewis肺癌C_(57)BL/6小鼠IFN-γ、TXB_2和6-酮-PGF1α影响的实验研究  被引量:3

摘  要:目的通过山仙颗粒对荷Lewis肺癌C57BL/6小鼠脾细胞IFN-γ活性及血浆中TXB2、6-Keto-PGF1α水平的影响,探讨其抗侵袭与转移的机理。方法将40只C57BL/6小鼠随机取出10只作为空白对照组(A组),把Lewis肺癌细胞接种于其余小鼠右腋皮下,将它们随机分成荷瘤对照组(B组)、金龙胶囊组(C组)、山仙颗粒组(D组)。接种24 h后,C、D组分别以金龙胶囊(50 mg/mL)、山仙颗粒(0.5 g/mL)悬液灌胃,A、B组用生理盐水灌胃,观察出瘤时间。造模第21天后处死小鼠,测移植瘤重、体质量、肺转移灶数及血浆中TXB2、6-Keto-PGF1α的水平。结果D组小鼠出瘤时间明显延长(P<0.01),瘤重及肺转移灶数均比B组低(P<0.01),而体重比B组高(P<0.01);D组小鼠脾细胞IFN-γ活性较B组高(P<0.05),且山仙颗粒能使血浆中TXB2降低,使TXB2/6-Keto-PGF1α的比值向不利于血栓形成方向变化。结论山仙颗粒有抑制肿瘤生长及抗转移的作用,其机理与增强宿主脾细胞IFN-γ活性及调节血浆中TXB2/6-Keto-PGF1α的比值趋于平衡有关。Objective To investigate the regulation of spleen IFN-γ, serum TXB2 and 6-Keto-PGF1α of C57 BL/6 inbred mice implanted Lewis lung cancer, and to explain the anti-metastasis of the Shanxian granule( SXG). Methods 10 C57BL/6 mice were selected from the 40 mice at random as blank group (A group). The rest 30 mice were planted by Lewis lung cancer cells. And they were divided randomly into Lewis lung cancer control group (B group) , JinLong capsule (JLC) group( C group) , and SXG group (D group). After planted 24 hours, the mice of C-group were given JLC (50 mg/mL), the mice of D-group were given SXG (0.5 g/mL). The mice of A, B groups were given normal saline. And we observed the time of the tumor coming into being. The 21 days after making model, we killed the 40 mice, and measured the weight of the transplanted tumor and mice, etc. Results The time of the tumor coming into being of D-group mice were obviously longer than that of B-group(P 〈 0.01 ). And the weight of mice were higher than that of B-group(P 〈 0.01 ). The activity of D-group. IFN-γ were higher than that of B-group. SXG can reduce the TXB2/6-Keto- PGF1α ratio. Conclusion SXG can inhibit the growth and metastasis of Lewis lung cancer and its possible mechanism is the enhancement of the activity of IFN-γ, and the reduction of the TXB2/6-Keto- PGF1α ratio.
