摘 要:目的探讨不同证型肺癌患者血清中蛋白质质谱的差异。方法用WCX2蛋白芯片结合表面增强激光解析电离飞行时问质谱(SELDI—TOF—MS)技术,分别检测12例气阴两虚肺癌和12例气滞血淤肺癌的血清蛋白质谱,筛选出差异表达蛋白质。结果共检测到113个有效的蛋白质波蜂,其中m/z位于2000~10000的波峰有104个,筛选出m/z为3952.05、3772.35、4170.07、3679.34、3156.38、4293.14的6个血清蛋白质。结论利用SELDI-TOF—MS技术将有可能建立新的中医肺癌辩证模式——蛋白质指纹图谱。Objective To discuss the difference of protein mass pectrometer between the Qi Yin Liang Xu Zheng patients with lung cancer and Qi Zhi Xue Yu Zheng patients with lung cancer. Methods We detected the protein mass pectrometer of 12 Qi Yin Liang Xu Zheng patients with lung cancer and 12 Qi Zhi Xue Yu Zheng patients with lung cancer with WCX2 protein combined with SELDI-TOF-MS technique, after that, we screened the different expressed protein. Results In this experiment we could detect 113 effective protein wave crests, and there were 104 wave crests which m/z located between 2 000 and 10 000. 6 serum proteins screened were m/z 3 952.05, 3 772. 35, 4 170.07, 3 679.34, 3 156.38, 4 293.14 respectively. Conclusion The SELDI-TOF-MS tecnique will be helpful to establish a new Chinese Medicine diagnostic model of diseases with proteomics and bioinfomatics and related techinque-protein fingerprint mapping.
