诺帝对肺腺癌细胞系A549细胞增殖的影响

摘　　要：目的探讨化学合成物诺帝对人肺腺癌细胞系A549细胞增殖的影响。方法采用自行合成的化合物诺帝(终浓度为100μmol/L)处理A549细胞,处理后12、24、48和72 h,用细胞培养、流式细胞仪检测、免疫组化及图像分析等方法检测诺帝对A549细胞的影响。结果诺帝可使A549细胞增殖受抑制,对细胞周期抑制作用主要表现于G1→S期;诺帝处理后,A549细胞增殖细胞核抗原(PCNA)表达明显减弱。结论诺帝对A549细胞的增殖有显著抑制作用,这种作用与其抑制PCNA有关。Objective To investigate the effects of Nordy synthesized in our laboratory on proliferation and apoptosis of human pulmonary adenocarcinoma cell line A549. Methods A549 ceils were treated with Nordy synthesized in our laboratory （whole concentration 100 umol/L）. In 12, 24, 48 and 72 hours, flow cytometry, immunohistoehemistry and image analysis were employed to detect the effect on the cell line A549. Results A549 cell proliferation was inhibited by Nordy in the G1→S phase. The expression of PCNA was downregulated in the ceils after treatmem by Nordy. Conclusion Nordy may play a significant role on proliferation of A549 ceil and this role may have a correlation with the reduced expression of PCNA.
