摘 要:目的为寻找一种较为简便经济实用的临床菌种保存方法,对纸片法与CRYOBANK菌种保存管法进行比较。方法将需保存的菌种纯化后分别用纸片法和CRYOBANK菌种保存管保存1、2、3、6、9、12个月后,进行复苏,比较存活率。结果苛氧菌用纸片法和CRYOBANK菌种保存管保存时,3个月后两种方法的存活率差异无统计学意义,保存6个月后两种方法的存活率差异有统计学意义;非苛氧菌用纸片法和CRYOBANK菌种保存管保存时,在各时间段两种方法的存活率差异无统计学意义。结论从经济实用的角度出发,非苛氧菌保存1年时适于用纸片法,而苛氧菌保存3个月内可用纸片法,保存6个月以上时必须用CRYOBANK菌种保存管。Objective To seek for a more simple, economical and practical method of preservation of strains by means of comparison of paper method and CRYOBANK preservative tube method. Methods The purified strains were preserved with paper method and CRYOBANK preservative tube method. At 1, 2, 3, 6, 9, 12 months timepoints, the preserved strains were revitalized, and their survival rate was determined and compared. Results There wasn't significant difference in survival rate of fastidous bacteria at 3 months time-point between the two methods, but the difference was evident at 6 months time-point. There weren't significant difference in survival rate of non- fastidous bacteria at all time-points preserved with two methods. Conclusion In the view of economy and practice, paper method may be adopted to preserve non-fastidous bacteria for one year and fastidous bacteria for 3 months. CRYOBANK preservative tube method should be employed to preserve fastidous bacteria for 6 months or longer.
