制备脐血LAK细胞的一种简单有效的方法

摘　　要：以健康脐带血为来源，采用３％明胶沉降法分离获取ＬＡＫ前体细胞，并在培养体系中增加植物血凝素诱导因子．结果：该法与常规淋巴细胞分层液分离法均能从脐带血中获取大量的ＬＡＫ前体细胞，且两者体外增殖情况及杀瘤活性并无明显差异（Ｐ＞０．０５）．提示新法简单快速，能从脐带血中制备出大量高抗肿瘤活性的ＬＡＫ细胞．With healthy umbilical blood as a source, pre LAK cells were acquired by using a method of 3% gelatin precipitation and added in the culture system were some inducing factors like phytohematoagglutinin (PHA). Result: a large number of pre LAK cells can be acquired from the umbilical blood by both gelatin precipitation and regular Ficoll Hypaque centrifugation methods, between which there was no marked difference (P>0.05) for external proliferation and killing activities. All this suggested that the new method, simple and speedy, can prepare a huge number of anti tumor activity LAK cells.
