摘　　要：目的观察CD81在正常大鼠神经视网膜上的表达。方法用抗CD81抗体对正常大鼠视网膜组织进行免疫组织化学染色,并对神经视网膜进行Western blot分析,同时用细胞免疫组织化学染色法检测体外培养视网膜神经胶质细胞的CD81表达。结果大鼠视网膜神经节细胞层、内丛状层和外丛状层呈阳性网状致密染色;Western blot分析神经视网膜层出现CD81阳性条带;体外培养的大鼠视网膜神经胶质细胞也呈CD81阳性表达。结论正常大鼠的神经视网膜表达CD81,视网膜神经胶质细胞是表达CD81的一种细胞类型。Objective As a member of tetraspanins superfamily,CD81 is proved to be involved in the inhibition of cellular mitotic activity,including rat retinal glial cells.The present study was to examine the distribution of CD81 in the normal rat neural retina.MethodsThe retina of Sprague-Dawley rat was isolated and rat retinal glial cells were cultured using Hicks method and identified by keratin and GFAP staining.Western blot analysis was performed to determine the expression of CD81 in the normal neural retina of Sprague-Dawley rats.The distribution of the protein on both rat neural retina and cultural retinal glial cells was assessed using immunohistochemistry.ResultsCD81 was found to express in the neural retina with a distinct laminar pattern of labeling.The CD81 positive staining existed throughout the ganglion cell layer,inner plexiform layer and outer plexiform layer in a dense reticular pattern.By Western blot analysis,the expression of CD81 was further confirmed.In cell culture,CD81 expression was located on the surface of retinal glial cells and concentrated at regions of cell–cell contact.Cultured and passaged retinal glial cells showed the positive reaction for GFAP and absent reaction for keratin.ConclusionCD81 could be expressed in the normal rat neural retina,especially on the membrane of retinal glial cells.
