摘  要:目的观察组蛋白去乙酰化酶抑制剂一曲古抑菌素(TSA)在体外和体内对胆囊癌细胞和肝外胆管癌细胞HDACl表达的影响。方法用TSA作用于胆囊癌细胞系(Mz-ChA-1)和肝外胆管癌细胞系(QBC939、KMBC、OZ),然后用逆转录.聚合酶链反应(RT.PCR)检测HDACl之mR—NA的表达变化,用Westernblot检测其蛋白的表达变化。将这些细胞接种在裸鼠皮下建立胆囊癌和肝外胆管癌裸鼠种植瘤模型,用免疫组织化学方法观察TSA在体内对裸鼠种植瘤组织中HDACl蛋白表达的影响。结果TSA可以减弱胆囊癌细胞系(Mz.ChA-1)和肝外胆管癌细胞系(QBC939、KMBC、OZ)HDAClmRNA和蛋白的表达;TSA作用前后的胆囊癌细胞系(Mz-ChA-1)和肝外胆管癌细胞系(KMBC)裸鼠成瘤组织中的各种蛋白的表达均无变化。结论TSA在体外可以抑制胆囊癌和肝外胆管癌HDACl的表达;TSA抑制HDACl表达的作用可能受到体内环境的影响而消失。Objective To study the effect of histone deacetylase inhibitor-trichostatin A (TSA) on the HDACl expression in gallbladder carcinoma and extrahepatic cholangiocarcinoma cells in vivo and in vitro. Methods The cells from gallbladder carcinoma cell line ( Mz-ChA-1 ) and cholangiocarcinoma cell lines (QBC939, KMBC and OZ) were treated with TSA, and the expression of HDACl mRNA and protein was detected by RT-PCR assay and Western blot respectively. These cells were subcutaneously transplanted into nude mice to establish the transplanted cholangiocarcinoma and gallbladder carcinoma models. The effect of TSA on the expression of HDACl protein in transplanted cancer tissues in vivo was observed. Results TSA could down-regulate the expression of HDACl mRNA and protein in gallbladder carcinoma cells and cholangiocarcinoma cells, TSA could not down-or up-regulate the expression of HDACl protein in the transplanted biliary tract cancer models in nude mice. Conclusion TSA might down-regulate the expression of HDACl in gallbladder carcinoma cells and cholangiocarcinoma cells in vitro,but this effect of TSA may disappear under the internal environment.
