摘 要:目的观察高脂喂养的胰岛素抵抗大鼠模型C反应蛋白(CRP)水平的动态变化;观察罗格列酮对肥胖大鼠血清CRP水平的影响。方法将190~240g健康清洁级近交系SD雄性大鼠36只,按完全随机法分为两组,正常对照组(NC组)和高脂饲养实验组,8周后胰岛素抵抗大鼠造模成功。将高脂饲养大鼠随机分为单纯高脂喂养组(HF组)、罗格列酮干预组(HFR组),于实验过程中取血检测空腹血糖、空腹胰岛素和血清CRP。结果经高脂饲料喂养8周后,模型组胰岛素敏感指数降低,与对照组比较,差异具有显著性(P<0.001)。经高脂饲料喂养8周后,模型组C反应蛋白水平明显升高,与正常对照组比较,差异具有显著性(P<0.001)。经过8周的干预治疗,HFR组与HF组比较,HFR组ISI显著增高(P<0.001),但仍低于NC组(P<0.001)。而HFR组经过干预后血清CRP均较为稳定,与HF组相比明显改善,差异有显著性(P<0.001),但而高于NC组(P<0.001)。结论随着胰岛素抵抗的加重,血清CRP水平逐渐升高,罗格列酮治疗可改善胰岛素抵抗,降低CRP水平。[Objective] To observe dynamic changes of insulin resistance, serum C-reaction protein levels in high-fed-diet rats with insulin resistance and effect of Rosiglitazone intervention. [Methods] 36 male Sprague Dawley rats were randomized to receive high-fat-diet and standard chow diet for 8 weeks, and feeding high-fat diet induced insulin resistance in rats. Consequently model rats were randomly divided into two groups: the HF group (high-fat diet only) and the HFR group (Rosiglitazone-treatment) fed with high-fat diet and Rosiglitazone [4 mg/(kg·d)] in a 8 weeks trial, the NC and HF group given tales doses Sodium Chloride as placebo. At the same time, the seru concentration of blood glucose, insulin, C-reactive protein (CRP), and bodyweight were measured. [Results] Rats fed with high-fat diet for 8 weeks resulted in loss of insulin sensitivity. It had significant difference compared with the NC group (P 〈0.001). Rats fed with high-fat diet for 8 weeks resulted in significantly higher level of CRP (P 〈0.001). After intervention study for 8 weeks, the ISI in HFR group was significantly higher than that of HF group (P 〈0.001), but compared to NC group, it still lower in ISI. And the CRP concentration of HFR group was much stable, but lower than that of HF group and higher than that of NC group (P 〈0.001). [Conclusion] The serum CRP con- centration gradually increased with worsening of insulin resistance.Rosiglitazonc improved insulin resistance,decreased the serum CRP levels.
