摘 要:美丽镰刀菌(Fusarium mairei)是一分离自南方红豆杉(Taxus chinensisvar.mairei)的产紫杉醇内生真菌,用B5培养基培养6 d,去菌丝后制得美丽镰刀菌培养液,并从中提取其胞外多糖.研究了4%(V/V)美丽镰刀菌培养液及4%(W/V)胞外多糖两种处理,对东北红豆杉(T.cuspidata)悬浮细胞防御反应及紫杉醇合成的影响.结果表明:两种处理均能诱导东北红豆杉细胞的防御反应.但美丽镰刀菌培养液的影响明显大于胞外多糖(P<0.05).另外,两种处理均可促进东北红豆杉细胞紫杉醇的合成与释放.美丽镰刀菌培养液处理得到的紫杉醇与其释放率分别是对照的2.5倍8.8倍,而胞外多糖处理得到的紫杉醇与其释放率则分别是对照的1.5倍与3倍.Fusarium mairei ,a taxol producing endophytic fungus, was derived from Taxus chinensis var. mairei. The F. mairei culture broth (FMCB) was prepared via filtering out the mycelia of F. rnairei cultured in liquid B5 medium for 6 days. And the exopolysaccharide (EPS) was abstracted from FMCB. The suspension cultures of T. cuspidata was treated with 4% FMCB (V/V) and 4% EPS (W/V) ,respectively. The results show that they all triggered the defense response of T. cuspidata cells. However,the efficiency of FMCB treatment was significantly higher than that of EPS treatment (P〈0.05). Additionally,they enhanced the taxol yield and release ratio of T. cuspidata cells. The taxol yields of cultures treated with FMCB and EPS were 2.5 fold and 1.5-fold more than that of control cultures,respectively,and the release ratios of taxol from FMCB-treated and EPS-treated cells were 8. 8 and 3 times more than that of control cells, respectively.
