摘  要:目的观察用于快速检测农药残留的大豆酯酶的稳定性,确定其最适保存条件。方法分别测定不同温度、不同酸碱度和不同保护剂对大豆酯酶酶活力的影响。结果在4℃保存30d,酶活力损失较少;在-20℃保存30d,完全失活;温度在50℃以下,pH在6.0~9.0保存10min,酶活力无明显变化;保护效果好的保护剂依次为牛血清白蛋白(BSA)、蔗糖和甘油。结论已确定大豆酯酶最适保存条件。Objective To study the stability of soybean esterase for rapid detection of residual pesticide and optimize the condition for storage. Methods Observe the influence of temperature, acidity and alkalinity as well as protective additive on activity of soybean esterase. Results After storage at 4℃ for 30 d, 80% of activity of soybean esterase remained. However, after storage at -20℃ for 30 d, the soybean esterase showed no activity The activity of soybean esterase showed no significant change after storage at pH 6.0 - 9.0, 50℃ below for 10 min. In the turn of protective effect, the protective additives were bovine serum albumin, sucrose and glycerol. Conclusion The condition for storage of soybean esterase was optimized.
