摘 要:[目的]探讨15-羟基前列腺素脱氢酶(15-hydroxyproglandin dehydrogenase,PGDH)基因甲基化状态与散发性结直肠癌发生发展的关系。[方法]利用甲基化特异性聚合酶链反应(methylation specific PCR,MSP)检测散发性结直肠癌及相对应的癌旁组织中PGDH基因甲基化状态。[结果]PGDH基因在散发性结直肠癌中甲基化率为37.5%(10/32),有6例癌及癌旁组织都发生了甲基化;伴有淋巴结及远处转移的甲基化率为50%(7/14),高于无转移的甲基化率16.7%(3/18)(P﹤0.05)。[结论]PGDH基因高甲基化是散发性结直肠癌中常见的分子改变之一,结直肠癌中PGDH基因高甲基化可能发生在癌变早期并与结直肠癌的恶性进展具有相关性。[ Objective] To explore the relationship between CpG methylation of 15-hydroxyproglandin dehydrogenase (PGDH) gene and histological features of sporadic colorectal cancer. [ Methods] Methylation-speeific PCR (MSP) was used to analyze the methyladon status of PGDH tumor suppressor gene in 32 sporadic colorectal cancers and corresponding normal tissues. [Results] Among the 32 colorectal cancers, PGDH hypermethylation was detected in 10 cases (37.5%), in the 10 methylated cases, 6 cases were happened hypermethylated in cancers and tissues adjacent to the cancer. 50% (7/14) of pa'tients occurred hypermethylation of PGDH gene and metastatic lymph node, which was significantly higher than those without metastasis (3/18, 16.7% ) (P 〈 0.05). [ Conclusion] Hypermethylation of PGDH gene is a frequent molecular event in sporadic colorectal cancer, it may occur at the early stage of the cancer and be related to invasion and metastasis of colorectal cancer.
