摘 要:目的研究1,25二羟维生素D3[1,25(OH)2D3]对大鼠肾小球系膜细胞生长及细胞周期的影响。方法通过肾小球系膜细胞体外培养技术,用含有不同浓度(0~10-6mol/L)1,25(OH)2D3的培养液干预系膜细胞,作用不同时间,应用噻唑蓝(MTT)法检测其对系膜细胞生长及增殖的影响,显微镜观察细胞形态变化,流式细胞仪(FCM)检测其对系膜细胞周期及凋亡的影响。结果0~10-6mol/L浓度的1,25(OH)2D3作用于系膜细胞48h均能显著抑制系膜细胞生长及增殖(P<0.01),并呈浓度依赖性;一定浓度的1,25(OH)2D3(10-8mol/L)对系膜细胞生长的抑制作用在一定时间范围内随着时间的延长,其抑制作用增强。不同浓度的1,(OH)2D3作用系膜细胞72h后,各组G0/G1期细胞含量均较对照组增高,S期含量随之减少,尤以10-6mol/L(P<0.01)、10-8mol/L(P<0.05)显著,并呈浓度依赖性,G2/M期含量无显著变化(P>0.05)。各干预组均可见亚G1凋亡峰。结论0~10-6mol/L浓度的1,25(OH)2D3能够抑制系膜细胞的生长及增殖,促进其凋亡,并呈浓度及时间依赖性。[Objective] To study the effect of 1, 25-dihydroxyvitamin D3 on the growth and cell cycle of cultured rat glomerular mesangial cells. [Methods] The cultured rat glomerular mesangial cells were treated with different dose of 1, 25(OH)2D3(0-10^-6 mol/L) for different time, the effect of mesangial cells proliferation were mesured by MTT colorimetric assay and the cell cycles were mesured by flow cytometry. [Results] After 48 h of 1, 25(OH)2D3(0-10^-4mol/L) incubation,the mesangial cells' growth and proliferation were inhibited significantly (P〈0.01) and showed dose-dependently. The inhibited effect of a certain concentration 1,25(OH)2D3(10^-8 mol/L) to mesangial cells showed time-dependently in definite range of time.After 72 h treatment of different concentration 1, 25(OH)2D3, significant difference were found in the cell cycle progression in response to 1,25 (OH)2D3 from the G1 to the S phases and apeptotic peak when compaired with thoses in the control group, especailly in the 10^-6mol/L (P〈0.01), 10^-4 mol/L(P 〈0.05) concentration. The G2/M phases had no significant changes(P 〉0.05). [Conelutions] 0-10^-6 mol/L 25 (OH)2D3 can inhibited the growth and proliferation of mesangial ceils and promote their apoptosis in a concentration-dependent and time- dependent manner.
