摘 要:目的观察维甲酸与人参皂甙-Rh2或半边旗二萜类化合物5F协同诱导肝癌细胞凋亡及对Fas蛋白表达的影响。方法体外培养肝癌细胞,设RA、GS-Rh2、5F、RA+GS-Rh2、RA+5F3个实验组和对照组,分别收集其药物作用24和48h后的培养细胞,应用流式细胞仪(FCM)检测肝癌细胞的凋亡率及Fas蛋白的表达。结果肝癌细胞药物作用48h时,RA+GS-Rh2组、RA+5F组与RA组或GS-Rh2组或5F组比较凋亡细胞数和Fas蛋白的表达有显著增高(Р<0.05),但在24h时,RA+GS-Rh2组、RA+5F组与RA组比较"亚G1峰"差异无显著性(Р>0.05)。结论维甲酸与人参皂甙-Rh2或半边旗二萜类化合物5F协同作用可以增强诱导肝癌细胞凋亡,促进Fas蛋白的表达。[Objective] To observe the apoptosis of Hepatoeellular carcinoma cells induced by Retinoic acid(RA) and Ginsenoside-Rh2 (GS-Rh2) GS-Rh2.5F, or 5F from Pteris semipinnata L(5F) and to be the influence of the expression of Fas. [Methods] To establish RA, RA and GS-Rh2, RA and 5F groups and control groups. Hepatocellu-lar carcinoma ceils were treated with RA, RA and GS-Rh2, RA and 5F for 24 h and 48 h. The apoptosis and the expression of Fas in Hepatocellular carcinoma cells were detected by Flow cytometry (FCM). [Results] Obvious apoptotic cells and expression of Fas were found when Hepatoeellular carcinoma cells were treated with the RA and GS-Rh2, RA and 5F for 48 h than in the RA(P 〈0.01). No obvious apoptotie cell of sub-G1 were found in 24 h(P 〉 0.05). [Conclusion] These results suggested that RA and GS-Rh2 or RA and 5F could enhance the apoptosis and expression of fas of Hepatocellular carcinoma cells.
