间歇游泳运动对大鼠下丘脑弓状核Fos蛋白表达的影响

摘  要:目的以大鼠间歇游泳训练为模型,观察其下丘脑弓状核Fos蛋白在6周游泳训练结束后不同时间点的表达及变化规律。方法将30只雄性SD大鼠随机分为对照组(n=5)和运动组(n=25),6周游泳训练结束后用免疫组织化学技术链霉素亲和素-生物素-过氧化物酶技术(SABC)法检测即刻,0.5,1,2,4h5个时间点弓状核Fos蛋白的表达情况,通过图像分析系统和统计学软件进行图像和数据分析。结果与对照组相比,运动组Fos蛋白表达在运动结束后0h明显增加(P<0.05),0.5h达到峰值(P<0.05),1h表达下降(P<0.05),2h再次升高(P<0.05),4h降至正常对照水平(P>0.05)。结论运动性应激可以迅速促进下丘脑弓状核Fos蛋白的活化。Objective To explore the activation and variation of Fos protein in arcuate nuclei of hypothalamus in different time in rats after six-week intermittent swimming exercise. Methods Thirty male Sprague-Dawley rats were randomly divided into control group ( n = 5) and swimming exercise group ( n = 25). The brains of swimming exercise group were removed at 0 h, 0.5 h, 1 h, 2 h and 4 h after six-week training. The expression of Fos protein in arcuate nuclei of hypothalamus was detected by immunohistochemical SABC techniques, and analyzed by imaging analyzing system and statistic system. Results Compared with control group, the expression of Fos protein in arcuate nuclei in exercise group increased at 0 h (P〈0.05), peaked at 0.5 h (P〈0.05), started to decrease at 1 h (P〈0.05), but increased at 2 h again (P〈0.05) ,and returned to normal levels at 4 h (P〉0.05). Conclusion Exercise stress can promote the expression of Fos protein in arcuate nuclei of hypothalamus.
