轻度认知障碍患者淋巴细胞蛋白磷酸酯酶-2A的变化

摘　　要：【目的】探讨轻度认知功能障碍(mild cognitive impairment,MCI)患者淋巴细胞蛋白磷酸酯酶-2A(protein phosphotase-2 A,PP-2 A)的变化。【方法】用32P放射性标记法检测了PP-2 A的活性,并用免疫印迹法检测不同病程MCI患者及正常对照组血淋巴细胞PP-2 A的表达。【结果】1年MCI患者淋巴细胞PP-2 A的活性(0.91±0.06)与对照组(1.02±0.08)比较有降低(P<0.05),2年MCI(0.83±0.07)和3年MCI(0.85±0.07)患者淋巴细胞PP-2 A的活性与对照组比较有明显降低(P<0.01,P<0.01),与1年MCI的比较虽有降低的趋势,但无统计学意义(P>0.05)。1年MCI组淋巴细胞PP-2 A的表达(0.95±0.06)与对照组(1.01±0.07)比较无明显降低(P>0.05),而2年MCI组(0.85±0.07)及3年MCI组(0.86±0.06)的PP-2 A表达与对照组相比明显降低(P<0.01,P<0.01),1年MCI组与2年MCI组的PP-2 A的表达无明显差别(P>0.05),与3年MCI组也无差别(P>0.05)。【结论】MCI患者淋巴细胞PP-2 A的活性及表达降低,对MCI的诊断可能有参考价值。[ Objective] To explore the alterations of PP- 2 A in lymphocytes in patients with mild cognitive impairment. [Methods] The activity of PP- 2 A was determined by 32 p-labelled in control group, 1 - year MCI group, 2 - year MCI group and 3 - year MCI group, and the expression of PP - 2 A was measured by Westernblot in these groups too. [ Results] The activity of PP - 2 A was decreased in 1 - year MCI group （0.91 ± 0.06） as compared with controls （ 1.02 ± 0.08） （ P 〈 0.05）. A lower decrease of PP- 2 A activity was also induced in 2 - year MCI group （0.83 ± 0.07） and 3 - year MCI group （0.85 ± 0.07） as compared with controls （P〈0.01, P〈0.01）o There was no obvious difference among the three MCI groups （P 〉 0.05）. In other hand, the expression of PP-2 A was not slgnificandy lower in 1- year MCI group （0.95 ±0.06） as compared with control group （1.01 ± 0.07）, but the expression of PP - 2 A in 2 - year MCI group （0.85 ± 0.07） and 3 - year MCI group （0.86 ±0.06） was lower as compared with control group （ P 〈 0.01, P 〈 0.01）. There was no obvious difference between 1 - year MCI group and 2 - year MCI group （ P 〉 0.05）, and no obvious difference was induced between 1 - year MCI group and 3 - year MCI group （ P 〉 0.05）. [ Conclusions] Lower expression and activity of PP- 2 A in lymphocytes implies a possible value of MCI diagnosis.
