摘  要:应用反相高效液相色谱-荧光检测技术,发展了人血清中14种胆汁酸分析的新方法,流动相中加入β环糊精作为添加剂,等度冲洗,一次分析可在60min内完成,且可以达到基线分离。由于胆汁酸的紫外吸收弱,采用4-溴甲基-7-甲氧基香豆素作为荧光试剂衍生。激发波长320nm,发射波长380nm。血清样品经预处理,回收率介于660%~916%之间。在0025~15mg/L质量浓度范围,色谱峰面积对浓度作线性回归分析,回归系数介于09908~09986之间。方法快速、简单、灵敏度高。A new HPLC method for the determination of free and conjugated bile acids in human sera has been developed Because of their weak UV absorption,bile acids are derivatized with the fluorescence reagent 4 bromomethyl 7 methoxycoumarin The excited wavelength is 320 nm,while emission wavelength is 380 nm β Cyclodextrin is used as an additive in the mobile phase Upon isocratic elution,baseline separation can be achieved and analyses can be completed within one hour Recoveries of human bile acids in serum sample varied from 66 0% to 91 6% and the RSD were 9 3%~21 5% for free and glycine conjugated bile acids The linear regression coefficients of 0 9908~0 9986 can be obtained,when peak areas are plotted versus injected amounts of free and glycine conjugated bile acids The method is simple,rapid,and sensitive
