摘 要:采用离子交换树脂对海藻糖提取液进行处理,以除去溶液中的杂质盐类及色素.首先经过初步静态筛选,从12种不同类型的树脂中筛选出吸附效果较好的8种树脂,再将树脂组成离子交换柱,然后经过层析柱动态筛选出3种树脂(001×7、D290、201×7),将筛选出的三种树脂组成串联柱,通过对串联式离子交换柱在不同操作条件进行优化实验,得到一组阳-阴-阴串联式离子交换柱(柱B:001×7(50mL)+D290(50mL)+001×7(20mL)),经柱B处理过后提取液的电导率从1700s/cm明显下降至40s/cm,离子交换柱处理量为4倍柱体积,吸光度为0.006,脱色率98%,海藻糖的收率为94%.Ion exchange resin was used to demineralize and decolorize from trehalose extract with ethanol. On the base of static adsorption, 8 resins were chosen from 12 different resins. Then, 3 resins were chosen from 8 resins from the chromatographic column on the base of dynamic adsorption. Column in series with 3 resins (001 × 7, D290, 201 × 7) was used to fulfill the dynamic adsorption experiments in different conditions. The optimum results showed that column B (001 × 7 (50 mL) +D290 (50 mL) +001 × 7 (20 mL)) was the best resin column. Electric conductivity oftrehalose extracting solvent decreased from 1700 μs/cm to 40 μs/cm. Absorbance of solvent decreased from 0.248 to 0.006 and decoloration rate was 98%. And the extraction recovery of trehalose was 94% with disposal volume 4 times of column volume.
