摘 要:[目的]研究Ca2+对水稻Ac/Ds愈伤组织增殖、分化及生根的影响。[方法]选用水稻品种Dongjin的Ac/Ds插入突变株系的种子,从成熟胚盾片诱导出愈伤组织,在筛选出的最适增殖、分化及生根培养基中添加不同浓度的Ca2+,研究Ca2+对水稻愈伤组织增殖、分化及生根的影响。[结果]适当增加Ca2+浓度能提高水稻愈伤组织的相对生长量、分化率、再生植株生根条数及根长,当Ca2+浓度为6.0mmol/L时效果最佳,当Ca2+浓度为7.5~15.0 mmol/L时水稻愈伤组织的相对生长量、分化率、再生植株的生根条数及根长呈下降趋势。[结论]适当增加培养基中Ca2+浓度可提高水稻Ac/Ds愈伤组织的相对生长量、分化率、再生植株生根条数及根长。[Objective] The study was to research the effect of Ca^2+ on proliferation,differentiation and rooting of callus of rice mutants with Ac/Ds.[Method] The seeds of rice mutants with Ac/Ds elements inserted from rice variety Dongjin were selected and the callus were induced from scutellum of mature embryo.Ca^2+ with different concn.was added into the selected optimum enrichment,differentiation and rooting medium to study the effect of Ca^2+ on proliferation,differentiation and rooting of rice callus.[Result] The relative growth rate and differentiation rate of rice callus and the rooting number and root length of regenerated plant could be increased when Ca^2+ concn.was increased properly,and the effect was better when Ca^2+ was 6.0 mmol/L,but the relative growth rate and differentiation rate of rice callus and the rooting number and root length of regenerated plant showed downward trend when Ca^2+ was 7.5-15.0 mmol/L.[Conclusion] The relative growth rate and differentiation rate of callus of rice mutants with Ac/Ds elements and the rooting number and root length of regenerated plant could be increased when Ca^2+ concn.was increased properly.
