缺血性脑卒中脑干单胺类神经递质的变化与卒中后睡眠障碍的研究  被引量:15

摘  要:搞要:目的利用大鼠中动脉栓塞模型,观察对比正常大鼠与中动脉栓塞24h后,大鼠脑干各区的去甲肾上腺素(NA)和多巴胺(DA)与五羟色胺(5-HT)含量的变化。方法18只健康成年雄性Spraque Dawley(SD)大鼠,随机分为正常组6只和模型组12只。其中模型组中6只用来取材测递质,6只做四氮唑染色(TTC染色)。模型成功后24h后,6只正常大鼠和6只模型大鼠断头取脑分离脑干,用高效液相色谱仪测递质。6只大鼠做TTC染色。结果显示大鼠在栓塞24h后,中脑的DA、NA、5-HT与正常组无明显区别。脑桥与延髓中DA、NA与正常组无明显区别,而5-HT明显高于正常组。与正常组对比P<0.05。结论单胺类递质在脑卒中后的变化过程,可能是一种动态平衡的破坏。是一种早期增高然后恢复到正常水平,进而降低的变化过程,而且不是单一递质的变化,而是多递质协同作用的结果。多递质的动态失衡(特别是递质减少阶段)促进了卒中后睡眠障碍的发生。Objective To utilize middle cercbral artery occlusion(MCAo )in rats,to observe and contrast the change of Noradrenaline(NA) ,Dopamine (DA) and serotonin(5-HT) in brain stem in normal rats and the middle cerebral artery(MCA) was surgically occluded after 24h in rats. Methods 18 healthy adult male SD rats were randomly divided into normal group(6) and model group (12). In the model group,six were use to measure transmittance, and six were use to tetrazolium color. Results After 24h of occlusion in rats,the DA,NA,5-HT in mesencephal had no obviously distinguish contrast that in normal rats. DA, NA in the bridge of varolius and medulla had no obvious distinguish :too,but 5-HT in the bridge of varolius and medulla was obviously higher than that in the normal rats(P〈0.05). Conclusion The change of momoamine neurotransmitter after stroke is possible a equilibrium being destroyed. It increase in the early,and then revive the normal level, and then decrease, it is not a single neurotransmitter's change. But many neurotransmitter in coordination with each other. Multitude neurotransmitter dynamic equilibrium is destroyed that lead to post-stroke sleep disorder.
