摘  要:为了揭示层粘连蛋白(laminin,Ln)信号传递作用与转移性肺腺癌增殖的关系。体外以外源性Ln(exogenousLn,exLn),抗Ln抗体(antibodyofLn,Ln-Ab)以及PMA[phorbol-12-myristate-13-acetate,蛋白激酶C(PKC)激活剂]作用于小鼠肺腺癌内源性Ln(endogenousLn,enLn)高表达的高转移克隆化正系(C6)及其低表达的低转移克隆化正系(C7)进行比较研究。结果:exLn能使C6细胞比C7细胞的增殖力在已是较高水平的基础上进一步提高,并与PMA的共同作用显示了增殖效应的协同。相反,Ln-Ab能使C7细胞比C6细胞的增殖力获得更明显抑制;并且,仅PMA的作用却使C6细胞比C7细胞的增殖力呈现更明显的抑制,并使C6细胞的跨膜Ca2+流恒态(homeostasis)发生显著性改变。结论:层粘连蛋白信号传递作用与高、低转移性小鼠肺腺癌细胞的增殖密切相关,这可能与PKC活性及跨膜Ca2+流恒态的变化相互关联.In order to explore the relationship of signal transductive effects of Laminin(Ln) onproliferation of mouse lung metastatic neoplasm. In vitro, effects of exopenous LA(exLn), antibody of Ln(Ln - Ab)and phorbol - 12 - myristate - 13 - acetate(PMA, protein kinase C(PKC) agonist) on the Proliferation of twocloned sttalns(C6/C7) were compared in an in vitro experiment. Both metastasis and endogenous Ln(enLn)expression were much higher in C6 cell than in C7 cell. Result: C6 cell possessal higher proliferative potentialthan C7 cell which was further increased by the aetion of exLn, and showed besides exLn in combination withPMA the synergistic action. The contrary, by the action of Ln- Ab, the Proliferation of C7 cell was decreaseddecmuch more than that of C6 cell. By the action of PMA alone, the proliferation of C6 cell was decreased muchmere than that of C7 cell and homeostasis of calcium ion across the plasma inelnblane(Ca2+ APM) of C6 cellwas also ehanged. Conclusion: The proliferation of the mouse long adenocarcinoma cells with low and highmetastasis were correlated to signal transductive effect of Ln, which might be related to the variation of bothPKC activity and Ca2 + APM homeostasis.
