摘  要:采用脑区微灌流技术结合以水杨酸为捕捉剂的IIPW-UV检测羟自由基方法和酶萤光检测谷氨酸方法,发现在大鼠纹状体细胞外灌流谷氨酸后,可使灌流液中的羟自由基显著增加,加灌氯胺酮可减轻谷氨酸的灌流效应。而灌流能产生超氧阴离子(O2)的黄嘌吟-黄嘌呤氧化酶系统,可使灌流液中的谷氨酸显著增加,给予维生素E则可消除这种效应。结果提示纹状体细胞外液中谷氨酸增多与氧自由基生成之间有相互促进作用。Based on the brain areas push-pull microperfusion system ,the cooperation between excessive glutamate and the formation of Oxygen ho radical in extraeellular corn^ntS of rat striatUIn was studied.The formation of hydroxyl ladleal was measured by HPLC with UV detector using salicylic acid as a trapping a gent which reacted with it yielding 2,5-dihydroxybenzoic acid.The results showed that the level of hydroxyl radical in perfusates was raised by perfusing glutamate,it could be reduced by perfusing ketamine ,the concentration of glutamate in perfusates was increased by perfusing xanthine and xanthine oxidase-superoxide anion production system,this effect could be vanished by Vit E. These results suggested that there is a vicious cycle between the excessive glutamate and oxygen free radical formation in the extracellular compartments of rat striatum.
