摘  要:在体外观察全反式维甲酸对人肺腺癌细胞株SPC-A1的增殖和DNA、RNA以及蛋白质合成的影响。当细胞在含有5μmol/L、10μmol/L和20μmol/L维甲酸的培养液中培养时,发现细胞生长均受到明显抑制。细胞在含有10μmol/L维甲酸的培养液中培养2天后,其3H—TdR和3H-亮氨酸掺入率明显下降,而培养延长至4天后,3H—TdR、3H—UdR和3H-亮氨酸掺入率都明显下降。通过流式细胞仪分析发现经过维甲酸处理6天后的细胞,其G1/G1期的细胞(67.1%)明显高于对照组(52.1%)(P<0.01),而S期细胞(19.6%)明显低于对照组(35.8%)(P<001)。In vitro influence of all-trans-retinoic acid (RA) on the growth and the syntheses of DNA. RNA and protein of SPC - A1 human lung adenoma cell line was studied. It was found that the growth of cell line was markedly inhibited when cells were cultured in 5μmol/L. 10μmol/L and 20μmol/L RA. After cells were cultured in 10μmol/L RA for 2 days, the 3H -TdR and 3H - Leu incorporation declined.But after cells were treated by RA for 4 days, the 3H - TdR, 3H - UdR and 3H - Leu incorporation all declined. By flow cytometry, the cell number in G0/G1 phase of cells treated by RA for 6 days (67. 1 %) was higher than that in G0/G1 phase of control cells (52.1 %) (p<0.01), the cell number of S phase (19.6%) lower than that in S phase of control cells (35.8%) (P< 0.01 ).
