摘  要:目的:探索血小板糖蛋白(Glycoprotein,Gp)IIb-IIIa复合体的基因多态性是否与中青年脑梗死有关。方法:选取住院年龄<59岁的脑梗死患者作为梗死组84例,门诊参加体检年龄相匹配者作为对照组67例,均提取空腹静脉血检测白细胞中DNA;对DNA中GpIIb和GpIIIa基因多态性的分析分别采用FQ-PCR反应和经聚合酶链反应(PCR)后,用限制性内切酶ScrFI酶切,2.2%琼脂糖凝胶电泳,紫外分析仪观察,比较2组间各指标的差异。结果:GpI-IbSer843等位基因的纯合子型与中青年脑梗死的发生相关(P<0.05);梗死组与对照组均为GpIIIaPlA1基因型。结论:在中青年脑梗死中有GpIIbIle-Ser843的基因多态性异常;而GpIIIaPlA1基因型无异常。Objective: To explore the relationship between the GpIIb-IIIa polymorphism and cerebral iniarction in young and middle-aged adults. Methods.. From 2002 to 2007, 84 patients with cerebral infarction and 67 control subjects were investigated. Their ages were less than 59 years old. Peripheral venous blood samples were drawn from all subjects after overnight fasting to extract genomic DNA. The GpIIb genotype was determined from leukocyte DNA by FQ-PCR. The GpIIIa genotype was determined from leukocyte DNA by PCR followed by ScrF I digestion. After 2.2 % agarose gel electrophoresis with ethidium bromide staining, the products were analyzed with ultraviolet analyzer. All statistical analy ses were performed by using SPSS 11.5. Measurement data were compared by means of the t test. The enumeration data were compared by means of the X^2 test. Results: Homozygosity for the Ser843 allele of the glycoproteln IIb was associated with increased risk of cerebral infarction (P〈0.05). All donors expressed the GpIIIa P1A1 phenotype. Conclusion: The GpIIb IIe-Ser843 gene potymorphism is abnormal in cerebral infarction among young and middle-aged adults, and the GpIIIa P1A1 phenotype is normal in the cerebral infarction.
