摘  要:目的观察高脂饮食大鼠血清抵抗素水平及下丘脑细胞因子信号转导抑制因子-3(SOCS-3)基因表达变化及其关系,探讨高脂饮食引发肥胖的发生机制。方法雄性6周龄SD大鼠16只。体质量80~100 g。随机分为基础饮食组和高脂饮食组,每组8只。单笼饲养,自由饮食饮水,明暗周期12 h,环境温度21~23℃,每周称体质量、测体长和尾长1次。大鼠饲养9周后,以乌拉坦(0.4 g/kg)深度麻醉,取心脏全血离心,采用ELISA法检测其血清抵抗素水平,葡萄糖氧化酶法检测其血清葡萄糖水平。取其下丘脑组织100~120 mg,用Trizol一步法提取总mRNA,经电泳显示28、18、5 S清晰条带,测定A260/A280>1.8。应用反转录聚合酶链反应(RT-PCR)检测其SOCS-3 mRNA表达水平。PCR产物经18 g/L琼脂糖凝胶电泳后,利用凝胶图像分析系统测定电泳条带密度值,以甘油醛-3-磷酸脱氢酶(GAPDH)为内参照,计算SOCS-3 mRNA相对水平,结果以SOCS-3条带占GAPDH条带密度百分率(%)表示。结果1.高脂饮食组大鼠体质量、体长、尾长及Lee′s指数明显高于基础饮食组(Pa<0.001);2.高脂饮食组大鼠血清血糖及抵抗素水平明显升高,与基础饮食组比较均有显著性差异(Pa<0.01);3.高脂饮食组大鼠下丘脑SOCS-3 mRNA表达水平显著高于基础饮食组(P<0.05);4.二组大鼠血清抵抗素水平与下丘脑SOCS-3 mRNA表达水平无明显相关性(Pa>0.05)。结论高脂饮食可导致大鼠血清抵抗素水平升高,进而诱导SOCS-3基因表达,在肥胖、抵抗素所致的胰岛素抵抗过程中SOCS-3可能发挥了重要的介质作用。Objective To observe the concentrations of serum resistin and changes of suppressor of cytokine signaling - 3 ( SOCS - 3 ) gene expression in hypothalamus of rats fed with high fat diet, and to explore the pathogenesis of obesity. Methods The male rats aged 6 weeks,weighted 80 - 100 g were allocated into 2 groups randomly,each containing 8 rats. The rats of high fat diet group were fed with high fat diet ,the rats of basic diet group were fed with basic diet. The rats were housed in cages individually at a temperature of 21 -23℃ ,maintained on a light -dark cycles of 12 hours,given free access to tap water and food. Body weight,body length,and tail length were recorded every week. After 9 weeks,the blood in heart of rats was collected during the depth of urethane (0.4 g/kg) anesthesia for the following research. The concen- tration of serum resistin and glucose were measured by enzyme - linked immunosorbent assay and glucose oxidase method respectively. The hypothalamus with 100 - 120 mg was removed,of which the total RNA was collected by using Trizol reagent,28 S,18 S and 5 S bands were all clear in electrophoresis picture and the ratio A260/A280 was more than 1.8. The level of SOCS - 3 gene mRNA expression was evaluated by reverse tran- scription polymerase chain reaction (RT- PCR). The PCR products were examined by electrophoresis with 1.8% agarose gel and observed by gelimage analysis system. The relative amount of SOCS - 3 mRNA was measured normalizing to the mRNA levels of glyceraldchyde 3 - phosphate dehydrogenase (GAPDH) and expressed as the percentage of density of SOCS -3 band with density of GAPDH band. Statistical analysis was performed using the SPSS 15.0 statistical software program. Results 1. The weight,body length,tail length and Lee's index of rats fed with high fat diet increased significantly than those of basic diet group( Pa 〈0. 0O1 ). 2. The rats fed with high fat diet showed of significantly increased serum glucose and resistin levels compared with those of rats
