摘  要:目的探讨中国汉族人5-羟色胺受体4个候选基因(5-HT2A受体基因、5-H2C受体基因、5-HT6受体基因、5-HL受体基因)与单相抑郁症的遗传关联性。方法应用聚合酶链式反应扩增技术与限制性片断长度多态性技术(PCR—RFLP)分析5-羟色胺受体4个候选基因多态性,对132例抑郁症患者和180名正常对照者进行关联分析;采用汉密顿抑郁量表(HAMD)评定病例组症状表型。结果(1)5.HT2A、5-HT1Dβ和5-Hk的基因型和等位基因频率在病例组和对照组中差异无显著性(P〉0.05)。(2)5-HT2C基因型在2组中分布差异有显著性(Х^2=7.0887,P〈0.05),但进一步按性别分层分布差异无显著性(P〉0.05)。(3)5-HT2A、5-HT2C和5-HT6病例组中HAMD各因子评分在三种基因型间分布差异无显著性(P〉0.05);5·HT1Dβ病例组HAMD各因子评分,其中日夜变化症状G/G基因型[(1.74±1.25)分]和T/G基因型[(1.61±1.06)分]分值均低于T/T基因型[(2.44±1.19)分],在绝望感症状群中T/G基因型[(7.09±1.93)分]分值为低于G/G基因型[(7.82±1.85)分]和T/T基因型[(8.33±2.22)分]。结论5-HT2C受体基因可能与中国汉族人单相抑郁症有关联,5-HT1Dβ受体基因T/T基因型可能是单相抑郁症患者日夜变化症状的危险因子,T/G基因型可能是绝望感症状群的保护性因素。Objective To investigate the association between the four eandidate serotonin receptor genes (5-HT2A receptor gene,5-HT2C receptor gene,5-HT1Dβreceptor gene,5-HT6 receptor gene ) and unipolar depression(UPD) in Han nationality of Chinese. Methods The ease-control association study was adopted to analyze assoeiation of the four candidate serotonin reeeptor genes polymorphism between 132 UPD patients and 142 normal controls using the polymerase ehain reaction and restriction fragment length polymorphisms (PCR-RFLP) teehnique, and evaluate the symptom phenotypes of eases applying the Hamilton Rating Seale for Depression (HAMD). Results ( 1 ) Neither genotype and allelies of 5-HT2A ,5-HT1Dβ and 5-HT6 gene showed significant differences between eases and controls (P 〉 0.05 ) . (2) The genotype of 5-HT2C gene were found to be of significant difference between the two groups (Х^2 = 7. 0887, df = 2, P 〈 0.05 ) , however, there were no significant difference in sex,respectively. (3) The three genotypes distributions of 5-HT2A ,5-HT2C and 5-HT6 gene in HAMD factors were no significant difference in ease group(P 〉 0.05), however, to 5-HT1Dβgene, the G/G genotype( 1.74 ± 1.25 ) and T/G genotype ( 1.61 ± 1.06 ) in diurnal variation symptom scores were lower than T/T genotype ( 2.44 ± 1.19) ,and the T/G genotype (7.09 ± 1.93 )in hopelessness symptom scores were lower than G/G genotype ( 7.82 ±1.85 ) and T/T genotype ( 8.33 ± 2.22) in HAMD factors. Conclusion The polymorphism of 5-HT2C receptor gene might be related with Han UPD, but the relationship that might be needed repetitive proof with enlargement of samples. The T/T geneotype of 5-HT1Dβ receptor gene polymorphism might be risk factor of diurnal variation symptom, and T/G genotype might be protective factor of hopelessness symptom groups of UPD.
