结直肠癌患者外周血中人端粒酶逆转录酶mRNA的表达及其临床意义

摘　　要：目的探讨结直肠癌患者外周血中人端粒酶亚单位hTERTmRNA的表达及其意义。方法以hTERTmRNA为靶基因运用逆转录聚合酶链反应(RT-PCR)法检测76例结直肠癌、22例良性大肠疾病患者和30例正常对照人群外周血中hTERTmRNA的表达。结果76例结直肠癌患者中有52例外周血hTERTmRNA表达阳性,阳性率为68.4%,22例良性大肠疾病患者中有1例外周血hTERTmRNA表达阳性,二者比较有显著性差异(0.001);30例正常对照人群外周血中均无hTERTmRNA表达。hTERTmRNA的阳性检出率与肿瘤分化程度和TNM分期密切相关,与其他因素无相关性。结论外周血细胞中hTERTmRNA的表达与肿瘤分期相关,有可能作为结直肠癌微转移的一个监测指标。Objective To evaluate the expression and clinical signifcance of human telomerase reverse transcriptase mRNA in peripheral blood of colorectal cancer patients. Methods Using hTERT mRNA as a target gene, the expression ofhTERT mRNA in 76 patients with colorectal carcinoma was examined by using RT-PCR technique. Meanwhile ,other 22 benign colorectal disease patient and 30 normal controls were examined. Results The positive rates of hTERT mRNA in the PB of colorectal cancer patients was 68.4%（52/76）. The positive rate was significantly higher than that in benign control （P〈 0.001）.All 30 healthy volunteers were negative for hTERT mRNA expression. The expression of the hTERT mRNAs significantly correlated with depth of tumor invasion, and TNM stage. Conclusion The expression ofhTERT mRNA in peripheral blood cells correlates with the pathological staging of colorectal cancer and it could be of potential use to monitor the micrometastases of tumor.
