摘 要:采用扫描电镜定性观测预处理效果,采用正交实验方法确定最佳酶解工艺条件,研究氨浸膨爆玉米芯试样的酶水解行为。结果表明,氨浸膨爆预处理方法,能够改善玉米芯试样的膨化裂解预处理效果,有利于后续酶水解,最佳工艺条件为pH=4.8,温度45℃,时间48h,底物浓度75g·L-1,酶用量60IU.g-1。玉米芯试样经过酶解可得还原糖0.5g/g以上。The enzymatic hydrolysis of corn cob pretreated by ammonia marinating-explosion dilatation is investigated by pretreated material microstructure observation with scanning electron microscopy(SEM), and the optimal condition determination of enzymatic hydrolysis through the orthogonal design. The results indicate that the AEM can dilatate and separate lignocellulose effectively. The optimal pretreatment condition of AEM is pH = 4.8, 45℃, 48h, 75g corn cob/L and 60 IU enzyme/g-corn cob. More than 0.5 g-sugar/g-corn cob can be obtained by enzymatic hydrolysis under optimal conditon.
