曲古抑菌素A对人胆管上皮癌细胞增殖及凋亡的影响

摘　　要：【目的】探讨组蛋白去乙酰化酶抑制剂曲古抑菌素A对体外培养的人胆管上皮癌细胞增殖及凋亡的影响。【方法】采用MTT法和软琼脂克隆形成试验观察细胞的生长增殖能力,采用TUNEL染色法观察细胞凋亡指数。【结果】曲古抑菌素A抑制胆管癌QBC939细胞的生长,促进其凋亡发生,且呈剂量依赖性。【结论】曲古抑菌素A可以有效地抑制人胆管癌QBC939细胞的增殖并诱导其凋亡。[Objective]To investigate the effects of histone deacetytase inhibitors-trichostatin A on the proliferation and apoptosis of human cholangiocarcinoma cell. [Methods]The cell proliferation ability was observed by MTT method and the test of colony formation in soft agar respectively. Apoptotic index （AI） was evaluated by TUNEL staining. [Results] Trichostatin A could inhibit the growth of human cholangiocarcinoma cell and induce the apoptosis in a dose dependent manner. [Conclusion] Trichostatin A can effectively inhibit the growth of human cholangiocarcinoma cell, and induce the apoptosis.
