摘 要:目的检测LYL1在髓系白血病细胞中所调控的可能靶基因。方法建立过度表达LYL1的白血病稳定细胞株,应用全基因组表达芯片测定其基因表达谱。结果初步筛选出可能受LYL1调控的靶基因或EST共444个,其中表达上调335个,表达下调109个。文中列出了表达明显上调和下调的部分转录因子、原癌基因、抑癌基因、细胞因子或受体、信号转导分子及细胞凋亡相关蛋白。值得注意的是,上调基因中包括与红系分化密切相关的转录因子LMO2与GATA1和许多原癌基因,包括RAS家族成员、EHM2、Pin1、PAX8、RAF-1、GRAP-2及WNT5A等;下调基因中包括抑癌基因p53等。结论研究可能受LYL1调控的靶顺序对于解释其在髓系白血病细胞的作用有重要意义。Objective To find out the target genes or sequences possibly regulated by transcription factor LYL1 in myeloid leukemia cells. Methods Gene expression profile of leukemia cells over-expressing LYL1 was analyzed with human whole genome cDNA chips. Results Four hundred and forty-four genes or expressed sequence tags (EST) were possibly regulated by LYL1, including 335 up-regulated and 109 down-regulated. Some up-regulated or down-regulated genes such as transcription factors, protooncogenes, tumor suppressor genes, growth factors or receptors, signal transduction molecules and apoptosis-related proteins were listed. Noticeably, up-regulated genes included LMO2 and GATA1, transcription factors involved in erythroid differentiation and many proto-oncogenes including RAS family members, EHM2, Pinl, PAXS, RAF-1, GRAP-2 and WNTSA. Down-regulated genes included tumor suppressor p53. Conclusion These results may help to explain the possible roles of LYL1 in myeloid leukemia.
