摘 要:目的研究丙烯酰胺(acrylamide)对黑腹果蝇遗传毒性及生殖系统的影响。方法经口喂饲Oregon-K雄蝇不同浓度的丙烯酰胺,染毒后从各浓度组中随机取出50只,分别与Basc雌处女蝇以1∶2的比例放在培养管中交配,2d后为第Ⅰ窝,将雄蝇取出后与另外2只处女蝇交配3 d后为第Ⅱ窝,再将雄蝇取出后与另外2只处女蝇交配3 d后为第Ⅲ窝。从每管子一代中随机取出20只肾型红眼雌蝇,分别与2只棒眼雄蝇交配,至F2代孵出并观察记录结果。结果每100 ml分别含丙烯酰胺0.5 mg、1.0 mg、2.0 mg的培养基喂饲3 d的Oregon-K雄蝇的生殖细胞均有明显的致突变现象。分窝(2,3,3 d)实验表明,丙烯酰胺对果蝇各阶段生殖细胞均有致突变作用,对精母细胞阶段损害较明显。结论丙烯酰胺对果蝇,具有明显的遗传毒性,须作进一步研究,以确定丙烯酰胺对人类的此种危害。Objective To study the genotoxicity of Acrylamide and its effect on reproductive system of Drosophila melanogaster. Methods Drosophila melanogaster of male Oregon-K were fed on different concentration of Acrylamide through oral cavity. Fifty Drosophila were selected form different concentration group randomly and mated with the female virgin Drosophila of Basc brand in the proportion of 1 : 2 in the test tubes. After 2 days the Drosophila were the first broods. Then the male taken from the first broods mated with another 2 female virgin Drosophila, after 3 days the drosophila were the second broods. The male taken again from the second broods mated with another 2 female virgin Drosophila, after 3 days the drosophila were the third broods. Twenty red eye reniform female Drosophila taken from the first filial generation in every tube mated with 2 bar eye male by each other. Observe and record the consequences until the second filial generation produced. Results The reproductive cell of male Oregon-K Drosophila that was fed by the 100 ml culture containing 0.5 mg, 1.0 mg and 2.0 mg of Acrylamide for 3 days had a definite mutagenic activity. Acrylamide had the same mutagenicity to the germ cell of different period (2, 3, 3 d), and affected the spermatocytes brood positively. Conclusion Acrylamide has a vital negative effect in genotoxicitv of Drosophila. The same consequence of human should be further studied.
