摘  要:本论文对卵白蛋白的分离与纯化、卵白蛋白铁的制备和表征进行了实验研究。首先采用不同饱和度的氯化钠和硫酸铵对卵白蛋白进行了定量盐析条件的试验,发现通过一次氯化钠或硫酸铵盐析和一次等电点沉降获得的卵白蛋白产品,蛋白纯度等于甚至优于Sigma公司提供的标准卵白蛋白,进一步的研究表明只有氯化钠盐析法获得的卵白蛋白才适合制备卵白蛋白铁复合物。接着,研究了制备卵白蛋白铁的工艺条件,探索了不同摩尔配比对所制得的卵白蛋白铁复合物铁含量的影响,初步探讨了不同结合比卵白蛋白铁复合物的性质,并用紫外可见吸收光谱和傅立叶转换红外光谱对卵白蛋白铁复合物进行了表征。这些实验研究为进一步开发新型廉价的补铁剂奠定了基础。Experimental studies were performed on the separation and purification of ovalbumin and the preparation and characterization of ovalbumin-ferric complexes. Firstly, the conditions for quantitatively salting-out of ovalbumin by different saturated degrees of sodium chloride or ammonium sulfate were tested. It was found that one time of NaCl or ( NH4 ) 2SO4 salting-out followed by one time of isoelectrie precipitation was suitable for obtaining the ovalbumin products with purity of equal to or even better than that of standard ovalbumin provided by Sigma,but only the ovalbumin obtained by NaCl salting-out plus isoelectric precipitation was suitable for preparation of the soluble ovalbumin-ferric complexes. Secondly,the suitable materiel ratios of ovalbumin to FeCl3 for preparation of the soluble ovalbumin-ferric complexes were studied and defined, and the physicochemical properties, including the iron content, isoelectric point, particle size and surface Zeta potential, as well as UV-Vis and FT-IR spectra of these new complexes were addressed. This empirical study provides a new way for preparation of new and cheaper oral iron supplements.
