摘  要:目的观察谷物发酵液对急性缺氧大鼠某些组织生化指标的影响,评价其抗缺氧效果,为研发新型抗缺氧药物提供科学依据。方法将Wistar大鼠随机分为常氧对照组、缺氧对照组和缺氧治疗组。缺氧治疗组每日以谷物发酵液(200 mg/mL)4 mL分2次灌胃,其他2组以同样方法灌以等体积蒸馏水,连续7 d。至第7 d灌胃1 h后,缺氧2组置于低压舱内海拔8000 m停留5 h后,迅速活杀取其全血、大脑、心肌、肝脏和后肢骨骼肌样品,按试剂盒方法测定其生化指标。结果缺氧治疗组的全血乳酸含量〔(16.99±4.55)mmol/L〕显著低于缺氧对照组〔(24.82±4.51)mmol/L〕(P<0.01),而接近于常氧对照组〔(15.76±3.09)mmol/L〕,其他组织的乳酸含量均呈现类似变化。缺氧治疗组的心肌ATP含量和骨骼肌糖原含量〔(66.05±7.63)nmol/μg、(1.75±0.56)mg/g〕则分别显著高于缺氧对照组〔(57.90±5.32)nmol/μg〕、(0.86±0.36)mg/g〕(P<0.05,P<0.01),但也接近于常氧对照组〔(64.85±6.28)nmol/μg、(1.11±0.45)mg/g〕,其他组织的ATP和糖原含量变化也类似。各组血糖浓度和心肌LDH的活力差异无统计学意义。结论谷物发酵液对缺氧动物的能量代谢有较明显的改善作用。Objective To evaluate the effects of fermented juice from grain on biochemical index in some tissues of rat exposed to acute hypoxia, so as to provide scientific basis for developing a new anti-hypoxia drug. Methods Wistar rats were randomly divided into normal control, hypoxia control, and hypoxia treated group. The animals in hypoxia treated grtmp were treated with fermented juice from grain(200 mg of dried content/mL) intragastrically 2 mL bid for 7 d. The animals in both control groups were treated with equal volume of distilled water and similar administration method. Hypoxia experiment was performed by placing the animals into a hypobaric chamber at a simulated altitude of 8 000 m for 5 h on day 7 after treatment. Then all the animals were killed rapidly, the samples of blood, brain, myocardium, liver and skeletal muscle were taken and their biochemical indexes were determined according to the methods offered in the relevant reagent kits. Results The content of blood lactic acid in hypoxia treated group[ ( 16.99 ± 4.55)mmol/L] was lower than that in hypoxia control group [ (24.82 ±4.51 ) mmol/L, P 〈 0.01 ], and near to that in normal control group [ ( 15.76 ± 3.09)mmol/L]. The contents of lactic acid in other tissues showed similar change. The contents of myocardium ATP and skeletal muscular glycogen in hypoxia treated group [ (66.05 ± 7.63 )nmol/μg, (1.75 ± 0.56)mg/g respectively] were higher than those in hypoxia control groups [ (57.90 ± 5.32)nmol/μg, P 〈 0.05; (0.86 ± 0.36)mg/g, P 〈 0.01 ] respectively, and also near to those in normal control group [ (64.85 ± 6.28) nmol/μg, ( 1.11 ± 0.45 ) mg/g ]. The change of ATP, and glycogen in other tissues was also similar to those in myocardium and skeletal muscle. No significant change of blood sugar, and myocardial LDH was found among three groups. Conclusion The fermented juice from grain could significantly improve the energy metabolism of hypoxic animals.
