肿瘤抑制基因RECK在骨肉瘤组织及细胞系中表达的初步研究

摘　　要：目的通过检测RECK基因在人骨肉瘤组织及人骨肉瘤细胞MG-63的高低转移亚系中的表达,探讨RECK基因在骨肉瘤发生及转移过程中的作用。方法采用免疫组化法检测18例骨肉瘤组织标本中RECK蛋白的表达,再分别应用RT-PCR方法检测细胞亚系中RECK mRNA的表达、明胶酶谱检测细胞MMP-2的活化水平、Matrigel侵袭实验检测肿瘤细胞侵袭能力。结果骨肉瘤组织中RECK蛋白的吸光度值为(0.31±0.07),明显低于正常骨组织(P<0.01),RECK的表达与肺转移、生存率有关(均P<0.05),而与患者年龄、性别、肿瘤大小、病理分级无关;高转移细胞组RECK mRNA的含量明显低于低转移组(P<0.01),而MMP-2活化水平、细胞侵袭力则明显升高(均P<0.05)。结论RECK基因异常低表达可能参与骨肉瘤侵袭、转移机制,RECK基因可能成为判断骨肉瘤患者预后新的分子标志物及肿瘤治疗的新靶点。Objective To detect the expression of RECK gene in osteosarcoma tissues and highly and low metastatic cell sublines of human osteosarcoma cell MG-63 and explore its possible roles in the occurrence and metastasis of osteosarcoma. Methods The expression level of RECK in 18 osteosarcoma tissues was detected by immunohistochemistry. RT-PCR, gelatin zymography and matrigel invasion assay were respectively used to evaluate the endogenous expression of RECK mRNA, MMP-2 activation ratio and invasive capacity in two osteosarcoma cell sublines. Results The absorbance （A） value of RECK protein in osteosarcoma tissues was 0.31± 0.07, which was significantly lower than that in normal bone tissues （P〈0.01）. RECK expression was significantly correlated with the pulmonary metastasis and survival rate （both P〈0. 05）, but not with age, sex, tumor size and pathologic grades. Highly metastatic cell group expressed significantly lower mRNA level of RECK than low metastatic cell group （P〈0.01）, but showed higher MMP-2 activation ratio and invasive capacity （both P〈0.05）. Conclusion The abnormal lowexpression of RECK may participate in osteosarcoma invasion and metastasis, and may be a new prognostic molecular marker for osteosarcoma and a new therapeutic target for tumor.
