摘  要:目的探讨牛磺酸对锰致大鼠海马神经毒性的影响。方法无特定病原体级SD雄性大鼠36只随机平均分成3个组:对照组、染锰组、牛磺酸预防性干预组(牛磺酸组)。对照组腹腔注射0.9%的生理氯化钠溶液;染锰组每天腹腔注射氯化锰溶液15 mg/kg,连续12周;牛磺酸组染锰的同时给予牛磺酸干预,染锰剂量和方法与染锰组一致,牛磺酸剂量为200 mg/kg,每周3次,连续12周。最后一次注射后24 h处死大鼠,切取海马,透射电镜观察各组神经元超微结构并检测丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力。结果①从第4周开始到实验结束,染锰组、牛磺酸组同期体重均比对照组低,差异具有统计学意义(P<0.05);染锰组与牛磺酸组相比,从实验第10周后染锰组体重显著低于牛磺酸组(P<0.05),各组动物同期平均进食量无明显差异;②透射电镜观察,对照组海马神经元形态规则,染锰组海马神经元发生凋亡的形态学变化,线粒体等细胞器严重受损。牛磺酸组接近正常海马神经元的形态,胞质内各细胞器形态、结构基本正常;③染锰组MDA含量比对照组明显增加(P<0.05),SOD活力显著下降(P<0.05)。牛磺酸组MDA含量与对照组比差异无显著性,且比染锰组显著下降(P<0.05);SOD活力比对照组低(P<0.05),与染锰组相比有所恢复,但差异无统计学意义。结论牛磺酸可以抵抗锰致海马神经元的凋亡,降低MDA产量,并使SOD活力有所恢复。Objective To explore the effects of taurine on the neurotoxicity induced by manganese in hippocampus of rats. Methods 36 SD rats were randomly divided into 3 groups. The control was injected i.p. with NS. The manganese chloride (MnCl2 ) group was injected with 15 mg MnCl2/kg daily for 12 weeks. The taurine group was injected with 200 mg/kg taurine solution for 12 weeks, 3 days a week. At the same time, the rats were injected with the same MnCl2 as the MnCl2 group. 24 hours after the last injection, the ultrastructural changes in hippocampal neurons were observed in each group by transmission electron microscope and the activity of SOD and the contents of MDA in hippocampus were detected. Results (1)Compared with the control, body weights of rats in MnCl2 and taurine group were decreased from 4th the week to the end of the experiment( P 〈 0. 05). Comparied between MnCl2 group and taurine group, there was no significant difference from the 1st week to the 9th week, while the body weight of the former group was lighter than that of the latter at the end of the 10th week( P 〈 0. 05 ). There was no significance among all groups in food consumption of rats ; (2) In the control group, the shape of hippocampal neurons was regular; while in MnCl2 group, the nuclei manifested typical apoptosis. The hippocampal tissue morphology in taurine group was almost similar to the control group ; (3) In MnCl2 group, the contents of MDA was distinctly higher than that of the control and the activity of SOD was decreased( P 〈 0. 05 ). The contents of MDA in tanrine group had no significant difference compared with the control, but obviously decreased compared with MnCl2 group(P 〈 0. 05). However, the activity of SOD in taurine group was much lower than that of the control group and had no significant difference compared with the MnCl2 group.
