摘  要:目的利用肝癌细胞株SMMC-7721比较IGF-ⅡP3脱氧核酶的活性.方法噻唑蓝比色法(MTT)检测4种脱氧核酶在不同浓度、不同作用时间、不同给予方法时的催化活性.结果DRz1的抑制活性相对较高(P〈0.05);脱氧核酶对靶基因随浓度增加抑制率相应提高,呈现一定的浓度依赖性;脱氧核酶24h的抑制率明显高于其他实验组(P〈0.05);转染给予法显著优于直接给予法(P〈0.05).结论脱氧核酶对靶基因的抑制率呈浓度、时间依赖性.Objective To investigate the DRz activity effect of IGF-ⅡP3 on apoptosis of hepatocarcinoma cells SMMC-7721. Methods The activity effects of DRz derived from the different concentrations,different times,different means and the inhibition ratio of four DRz by MTT assay on apoptosis of hepatocarcinoma cells. Results The inhibitive activity of DRz1 was higher than others(P〈 0.05) ;The inhibitive ratio of the target gene were raised with the concentration of DRz increasing,and presented concentration dependence;The inhibitive activity of DRz for 24h was higher than others(P 〈 0.05). The method of transfection was better than the method of straight (P 〈 0.05). Conclusion The inhibitive ratio of the target gene relied on concentration and time.
