摘 要:目的:研究针刺任脉和督脉经穴对局灶性脑缺血大鼠缺血海马星形胶质细胞的调节作用。方法:100只成年健康雄性Wistar大鼠,随机分为假手术(sham)组、模型组、针刺督脉组及针刺任脉和督脉组,sham组10只,其余3组又分别随机分为7d组、14d组和28d组3个亚组,每亚组10只。Wistar大鼠以线栓法制作大鼠大脑中动脉栓塞(MCAO)模型。通过免疫荧光双标法研究各组胶质纤维酸性蛋白(GFAP)阳性细胞和胶质纤维酸性蛋白/神经元特异性烯醇化酶(GFAP/NSE)双标细胞的表达。结果:针刺督脉治疗可以下调脑缺血后海马齿状回的GFAP阳性细胞数,同时使GFAP/NSE双标细胞增多;针刺任脉和督脉治疗的GFAP阳性细胞增殖幅度小于针刺督脉组,而GFAP/NSE双标细胞的增殖幅度则大于针刺督脉组。结论:针刺任脉和督脉可抑制脑缺血后海马星形胶质细胞的过度增殖并可促进细胞分化。Objective: To investigate the regulatory effects of electroacupuncturing at Ren and Du vessels on hippocamp horizontal cells of focal cerebral isehemia. Methods: 100 Wistar rats were randomly divided into four groups: sham group, MCAO group, electroacupuncturing at Du vessel group and electroacupuneturing at Ren and Du vessels group, the sham group had 10 rats, the other three groups were further divided into three subgroups: 7 d group, 14 d group and 28 d group, with each subgroup of 10 rats, Wistar rats were employed to prepare the middle cerebral artery occlusion (MCAO). Then im munofluorescence double-staining method was applied to detect the expressions of glial fibrillary acidic protein (GFAP) positive cells and glial fibrillary acidic protein/neuron specific enolase (GFAP/NSE) double-immunostaining positive cells. Re- sults: Eleetroaeupuncturing Du vessel could down regulate the number of GFAP positive cells of the hippoeampal dentate gyrus in focal cerebral ischemia, and increased the number of GFAP/NSE double-immunostaining positive cells. The proliferation of GFAP positive cells in electroacupuneture at Ren and Du vessels group was fewer than that in the group of single elcctroaeupuncture at Du vessel group, and the proliferation of GFAP/NSE double-immunostaining positive cells was more than that in the electroacupuncture at Du vessels group. Conclusion: Electroacupuneturing at Ren and Du vessels can inhibit the over proliferation of hippocamp horizontal cells of focal cerebral isehemia and promote the cell differentiation.
