急性脑梗死大鼠COX-2表达及逐痰通络汤的干预作用

摘　　要：目的:通过观察脑梗死后环氧合酶-2(COX-2)的表达及神经功能,探讨中药逐痰通络汤对脑缺血损伤的保护作用。方法:清洁级雄性SD大鼠54只,随机分为假手术组、对照组、药物组。应用线栓法建立大脑中动脉栓塞(MCAO)模型,参考Bederson 6级5分法进行神经功能评分,免疫组化检测COX-2的表达。结果:①神经功能评分第3天达到高峰,第14天最低,第3天后药物组与对照组比较,差异有显著性意义(P<0.05)。②脑组织COX-2在术后第1天大量表达,第3天仍表达较高,但第7、14天表达极少。与对照组比较,药物组阳性细胞表达强度在1天、3天减少,差异有显著性意义(P<0.05)。结论:逐痰通络汤对急性局灶性脑缺血引起的脑损伤具有一定的保护作用,其保护机制可能与抑制炎症反应,减少COX-2的表达,抑制其神经毒性作用有关。Objective： To observe the expression of Cyclooxygenase-2 （COX-2） and the antagonism function of Zhutan Tongluo decoction in acute cerebral infarction of rats. Methods：54 sprague-Dawley male rats were divided randomly into the sham operation group, the contrast group and the medicine group. The cerebral artery occlusion model was established by the thread-tying method. The score of nerve function were evaluated by the reference of Bederson＇s 6-class and 5-score. The expression of COX-2 was observed by immunohistochemical detection. Results： 1. score of nerve function ： the score at the 3^nl day was highest and that at the 14^th day lowest. There was significant difference between the medicine group and the contrast group after 3 day （P 〈 0.05 ）. 2. expression of COX-2 ： COX-2 was evidently expressed at the 1^at, day and also highly expressed at the 3^nl day, while rarely expressed at the 7^th and 14^th day. The expression in the medicine group at the 1^nl day and the 3^nl day was much decreased compared with that in the contrast group. The difference had significantly statistic meaning （ P 〈 0.05 ）. Conclusion ： Zhutan Tongluo decoction has some protection function on acute focal cerebral infarction of rats. Its protective mechanism maybe concerned with refraining inflammatory reaction, decreasing the x expression of COX-2, and neurotoxic effect.
