摘  要:目的探讨pcDNA3.1+Ape1质粒转染对血管内皮细胞电离辐射敏感性的影响。方法以pcDNA3.1+空载体转染、未转染细胞作对照,在细胞转染pcDNA3.1+Ape1质粒后48h,给予3组细胞2、4、6和8Gy高能x线照射,采用碱性单细胞凝胶电泳和克隆形成实验,分析在不同辐射剂量条件下pcDNA3.1+Ape1质粒转染对人脐静脉内皮细胞辐射敏感性的影响。结果与未转染细胞相比,3μgpcDNA3.1+Ape1质粒转染细胞的初始OTM和残存OTM值在2Gy照射时显著降低(P〈0.01),但8Gy照射时差异无统计学意义(P〉0.05);SF2显著增高(P〈0.05),但SF4、SF6及SF8的差异均无统计学意义(P〉0.05)。结论pcDNA3.1+Ape1质粒转染可增加内皮细胞对低剂量电离辐射的抵抗能力。Objective To investigate the possible effects on radiosensitivity in human umbilical vein endothelial cells after transfection of pcDNA3.1 + Ape1 plasmid. Methods The expressing vector peDNA3.1 + Ape1, the control vector pcDNA3.1 + or non-transfection ceils was irradiated by 2, 4, 6, and 8 Gy photon beam at 48 h post-transfection. The value of initial and residual Oliver tail moment (OTM) under the alkaline single cell gelelectrophoresis assay and the colony forming test were utilized as the markers for the evaluaton of cells intrinsic radiosensitivity. The effect on radiosensitivity in human umbilical vein endothelial ceils after transfeetion of the expressing vector pcDNA3.1 + Ape1 was analyzed according to the radio-dose, compared to the empty veeor control and non-transfection cells. Results The initial and residual OTM value of endothelial cells transfected by 3 μg pcDNA3.1 + Ape1 plasmid was lower significantly than ones of endothelial ceils untransfected at 2 Gy irradiation (P 〈 0.01), but was no significant difference at 8 Gy (P 〉 0.05), and SF2 was higher remarkably in transfeeted cells than one in untransfected cells ( P 〈 0.05), but SF4 , SF6 and SF8 were no significant differences (all of P 〉 0.05). Conclusions The transfection of pcDNA3.1 + Ape 1 plasmid could enhance radioresistance of endothelial cells to the low-dose irradiation.
