摘 要:目的研究番茄红素对去卵巢大鼠体内骨质的影响及其可能的作用机制。方法将32只雌性SD大鼠随机分成4组,分别为假手术(Sham)组,去卵巢(OVX)组,OVX+低L组,OVX+高L组。后两组于术后1w开始用番茄红素灌胃,10w后处死各组大鼠,并对血清钙、磷、碱性磷酸酶、MDA、SOD和骨密度等进行检测,对椎体骨行HE染色及NF-κB免疫组化染色。结果(1)与假手术组相比,模型组大鼠血清钙、磷、碱性磷酸酶、MDA等均不同程度的升高(P<0.01,P<0.05),而血清SOD水平、骨密度值指标均不同程度的下降(P<0.01,P<0.05)。相比之下,药物干预组尤其是大剂量组的上述参数指标与模型组相比呈现出显著差异(P<0.01,P<0.05)。(2)模型组与假手术组相比,NF-κB的表达灰度值升高(P<0.01),骨小梁萎缩变细、间隙增宽;干预组与模型组相比,NF-κB的表达灰度值不同程度降低(P<0.05,P<0.01),骨小梁增粗、间隙变小。结论初步证实番茄红素对去势大鼠骨质具有一定的保护作用,且这种保护作用可能与减少氧化应激和降低NF-κB的表达有关。Objective To observe the influence of lycopene on bone quality of ovariectomized rats, and to study the possible mechanism. Methods 32 female SD rats were randomly divided into 4 groups: sham operation (sham) group, ovariectomized (OVX) group, OVX treated with small dose lycopene group, OVX treated with large dose lycopene group. Bilateral ovariectomy was done on OVX group and OVX treated with lycopene groups. Therapeutic groups were treated with lycopene 1 week after operation. All rats were sacrificed in 10 weeks after operation, the serum calcium, phosphonium, alkaline phosphatase, malondialdehyde,SOD and bone density were tested, and the histomorphology of bone was disposaled by H-E staining and the NF-κB expression by immunohistochemistry staining. Results (1)The serum calcium, phosphonium, alkaline phosphatase and malondialdehyde of OVX group were higher than sham group (P 〈 0.01, P 〈 0.05). The SOD in serum and bone density of OVX group were lower than sham group ( P 〈 0.01, P 〈 0.05). All parameters referred in therapeutic groups presented significant differences compared with those in sham group( P 〈 0.01, P 〈 0.05), large dose lycopene therapeutic group especially. (2)The gray scale of NF-κB expression was higher in OVX group than sham group, and the bone trabeculae in OVX group became thinner and gaps became wider; compared with OVX group, the gray scale of NF-κB expression was lower in therapeutic groups. Conclusions We consider that lycopenc could protect OVX rats from osteoporosis, and this protection is related with the reduction of oxidative stress and the expression of NF-κB.
