摘  要:目的:探讨次级淋巴组织趋化因子(SLC)及其受体CCR7与血管内皮生长因子-C(VEGF-C)在胰腺癌组织中的表达及其与临床病理的关系。方法:应用免疫组化、RT-PCR和实时荧光定量PCR技术检测30例胰腺癌组织、癌旁组织、正常胰腺组织和胰周淋巴结组织中SLC、CCR7和VEGF-C的表达情况。结果:SLC蛋白在胰腺癌组织中呈低表达状态,在正常胰腺组织、癌旁组织和胰周淋巴结均呈中等表达状态,与胰腺癌组织中的表达相比较差异均有统计学意义,P<0.01。RT-PCR和实时荧光定量PCR结果也证实CCR7在胰腺癌组织、癌旁组织以及胰周淋巴结均呈高表达状态,而在正常胰腺组织呈低表达状态,P<0.01。经Spearman秩相关检验,胰腺癌组织中CCR7与VEGF-C蛋白的表达具有显著的相关性,P=0.038。结论:SLC的表达与胰腺癌淋巴结转移和TNM分期密切相关,并可能参与胰腺癌的淋巴管生成和淋巴结转移的调控。CCR7与VEGF-C的阳性表达可能对胰腺癌的发展起共同促进作用。OBJECTIVE: To investigate the expression of sec ondary lymphoid tissue chemokine (SLC), CCR7 and VEGF-C in pancreatic cancer and relation to clinical pathology. METHODS: The tissue samples including PAC, the cancerous peripheral tissues, the normal pancreatic tissues and peripheral lympho nodes were obtained from 30 patients with PAC. The expressions of SLC, CCRTand VEGF-C proteins in these tissues were assayed by immunohisto chemical staining. The expression of SLC, CCR7 and VEGF C mRNA in PAC were also investigated by reverse transcription polymerase chain reaction (RT-PCR) and fluorescence quantitative real time PCR. RESULTS: The positive rates of SLC protein in PAC was sig nificantly lower than those in cancerous peripheral tissues, normal pancreatic tissues and peripheral lympho nodes(P〈0.01), the expression levels of CCR7 mRNA in PAC tissues, the cancerous peripheral tissues and peripheral lympho nodes were higher than that in the normal pancreatic tissues, P〈0.01. There was significant correlation between the expressions of CCR7 and VEGF C protein in PAC which was tested by Spearman correlation test, P=0. 038. CONCLUSIONS: The SLC maybe have diphasic action of anti-tumor effect and promotion lymphatic metastasis during the development of PAC. The ex pression of CCR7 was significantly associated with lymphatic metastasis and TNM stages of PAC, and CCR7 maybe participate in the regulation of lymphangiogenesis and lymphatic metastasis of PAC. CCR7 and VEGF-C maybe promote the development of PAC mutually.
