食管鳞癌组织垂体瘤转化基因表达及其临床意义的研究

摘　　要：目的:探讨食管鳞癌组织中垂体瘤转化基因(PTTG)mRNA及其蛋白的表达及与肿瘤临床病理特征的关系。方法:采用RT-PCR技术检测30例食管鳞癌及相应的30例癌旁组织中PTTGmRNA的表达,同时采用免疫组织化学SP法检测标本中PTTG蛋白的表达。结果:在食管鳞癌组织中PTTGmRNA的阳性表达率(93.3%)及平均表达水平(0.69±0.06)显著高于相应的癌旁正常组织(70.0%和0.32±0.06),两组比较差异有统计学意义,P<0.01。PTTG蛋白在食管鳞癌组织中的阳性表达(83.3%,25/30)明显高于在癌旁组织中的阳性表达(20.0%,6/30),P<0.01;并与淋巴结转移、TNM分期和分化程度有关。结论:PTTG基因异常表达可对食管鳞癌的发生和发展起重要作用,可能为食管鳞癌的早期诊断和基因治疗提供新的靶点。OBJECTIVE： To investigate the expression and clinical significance of pituitary tumor-transforming gene （PTTG） in esophageal cancer. METHODS： Expression of PTTG mRNA and protein were detected by semi quantitative RT-PCR and immunohistochemistry respectively in 30 case of esophageal cancer and their adjacent normal esophageal tissues. RESULTS： The expression rate and level of PTTG mRNA in esophageal cancer were 93.3% and 0.69±0.06, which were significant higher than those in their adjacent normal esophageal tissues （70. 0% and 0.32± 0.06）. The expression of PTTG protein in esophageal cancer （83.3% ,25/30） was significantly higher than that in corresponding normal esophageal tissue （20%. 0,6/30）. The expression of PTTG was related to surgical-pathological stage, myometrial infiltration depth, lymphatic metastasis and pathological subtype （P〈0.05, respectively）, but was irrelevant to patients＇ age and pathological grade （P〉0.05, respectively）. CONCLUSION： PTTG may play an important role in carcinogenesis and development of esophageal cancer.
