摘　　要：目的:研究铜绿假单胞菌(pseudo-monas aeruginosa,PA)注射液在体外实验中对胃癌细胞的杀伤作用。方法:采用MTT法观察PA注射液抑制胃癌瘤株MKN45细胞增殖和杀伤作用的剂量依赖性;采用扫描电镜、透射电镜和TUNEL技术观察PA注射液与胃癌瘤株MKN45的特异性结合、致超微结构变化和诱导凋亡效应。结果:PA注射液能与胃癌瘤株MKN45细胞发生特异性结合并可进入细胞内,抑制其增殖和生长,其杀伤作用随着用药浓度增加而加强;PA注射液可以导致肿瘤细胞出现数量减少、体积缩小(细胞固缩)、细胞膜结构破坏、微绒毛减少、胞质内空泡、细胞核深染及核边聚等典型的细胞凋亡表现。结论:PA注射液能直接杀伤胃癌瘤株MKN45细胞,且这种杀伤作用存在剂量依赖性;其对胃癌瘤株细胞有抑制增殖和诱导凋亡的作用。OBJECTIVE：To evaluate the killing effect by PA-MSHA vaccine on gastric cancer cells in vitro.METHODS：MTT method was used to observe the inhibition and killing effect by PA-MSHA vaccine on gastric cancer cell strain MKN45.Specific combination,inhibition of cell proliferation,changes of super-microstructure and apoptosis of MKN45 were observed by transmission electron microscopy,scanning electron microscopy and TUNEL technique.RESULTS：PA-MSHA vaccine could specifically combine and penetrate into the gastric cancer cell strain MKN45 so as to inhibit its proliferation and the killing effect was in direct proportion to its dose.PA-MSHA vaccine could induce the decreasing in number and size,destruction of cell membrane and microvilli,vacuole in cytoplasm and deep staining of nucleus.CONCLUSIONS：PA-MSHA vaccine has dose-dependent killing effect on gastric cancer cell strain MKN45 by specific combination.It can also inhibit the proliferation of gastric cancer cell strain MKN45 and induce its apoptosis.
