摘  要:以芦丁为对照品,采用分光光度法测定叶下珠中总黄酮含量,在510nm处建立标准曲线,回归方程为A=10.649C-0.00557,其相关系数R=0.9982。采用正交试验设计探讨以乙醇为溶剂,从叶下珠中提取总黄酮的最佳工艺条件,以总黄酮得率为指标,考察乙醇浓度、提取温度、乙醇用量以及提取时间等4个因素对萃取效果的影响,结果表明:在本试验范围内,采用乙醇-水体系提取叶下珠中总黄酮类物质的最佳工艺条件为:70%的乙醇、浸提温度为90℃、料液比为1∶20、提取时间为1h,在此条件下,总黄酮得率为1.394%。Rutin as reference samper, the total flavonoids content in Phyllanthus urinaria L. was determined by spectrophotometry, establishing working curve in 510 nm.The regression equation of the standard curve was A =10.649C-0.005 57,and the correlation coefficient R was 0.998 2. The optimized process conditions for ethanol that used as solvent extracting total flavonoids from phyllanthus urinaria L were explored by orthogonal test. Based on the total flavonoids that is regarded as the objective, the influences of solvent concentration, extraction temperature, solvent dosage and extraction times on the extraction efficiency were studied. The results show that the optimized extraction conditions is the 70 % aqueous solution of ethanol, the volume of liquid(mL) ratio to the solid mass (g) is 20:1, the extraction temperature is 90 ℃, and extraction time is 1 h. Under this condition, the yield of total flavonoids is 1.394 % .
